Alcian Blue Stain Kit, pH 2.5 is a histochemical staining system for the selective demonstration of acidic mucopolysaccharides and acid mucins in tissue sections. It produces a characteristic blue coloration of acidic mucin and is commonly used in histopathology for the evaluation of mucin-producing tissues and pathological changes associated with mucin accumulation.

Working Principle At pH 2.5, Alcian Blue is a cationic copper-containing phthalocyanine dye that binds electrostatically to negatively charged acidic groups present in acidic mucins and related acidic mucopolysaccharides. These components are consequently demonstrated in blue, while the selected counterstain provides contrast for the surrounding tissue structures.
