Gill I Hematoxylin Solution
- Our Gill I Hematoxylin is a progressive, aluminum-based nuclear stain formulated as per Dr. G. W. Gill’s original formula (1974). It is the lightest strength in the Gill series – specifically designed for Gynecological Cytology (PAP Smears), Non-GYN Cytology, and Thin Biopsies.
- Unlike Harris Hematoxylin (regressive stain that overstains and needs acid differentiation), Gill I is ‘progressive’ – it stains nuclei to the precise endpoint and STOPS. No differentiation or bluing in acid-alcohol is required. This means nuclear chromatin details are never washed out.
- It is the most preferred Hematoxylin in the world for PAP stain and recommended by WHO for Cervical Cancer Screening Programs.
- Gill I vs Gill II vs Gill III :

1. Gill I : Single strength Hematoxylin, Best for Cytology PAP Smears, FNA, Body Fluids. Gives light, transparent nuclear stain so Pap chromatin is crystal clear. Preferred by cytotechnologists.
2. Gill II: Double strength of Gill I For Routine H&E Histology.
3. Gill III: Triple strength, Very strong, for urgent biopsies / when staining time needs to be 30 seconds only.
- True Progressive Stain: No differentiation with 1% Acid-Alcohol needed. This eliminates the #1 error of junior technicians – over-differentiation which causes faded nuclei and repeat staining.
- Optimized for PAP & Cytology: Low Hematoxylin concentration gives transparent nuclear staining. Chromatin pattern, intranuclear inclusions, mitosis are clearly visible – critical for cancer diagnosis. Harris is too opaque for PAP.
- Chemically Ripened, Ready-to-Use: lot-to-lot same intensity. No waiting.
- No Metallic Scum: No surface scum, no precipitate, no need to filter daily. Bottle lasts till last drop.
- IHC Compatible: Since it does NOT use Mercuric Oxide (unlike old Harris formulas), it does NOT inhibit subsequent IHC. You can do H&E, then destain and do IHC on same slide if needed.

1. Progressive Aluminum Lake Mechanism:
- Step 1 – Ripening: Hematoxylin itself is not a dye. It is naturally oxidized to Hematein, in this solution (chemically ripened, not sun-ripened – so lot-to-lot consistency is same).
- Step 2 – Lake Formation: Hematein + Aluminum Sulfate forms a blue-violet Aluminum-Hematein Lake (positively charged complex).
- Step 3 – Progressive Staining: This positively charged lake binds selectively to negatively charged nuclear DNA (Phosphate backbone). Since Gill I has low dye concentration, it saturates the nucleus slowly and reaches optimal intensity in 2-3 minutes and stops. No overstaining.
- Step 4 – Blueing: After water wash, color changes from red-purple to crisp blue when pH is increased to >5.0 (tap water blueing is enough, no need for Ammonia or Lithium Carbonate).

1. Beginner-Friendly: New technicians cannot over-stain with Gill I. If you forget slide for 5 minutes, it will still be good (with Harris, slide is black). Reduces repeat staining by 90%.
