  • Acid-fast bacilli possess a lipid-rich, waxy cell wall containing mycolic acids, which enables them to retain the primary stain during acid-fast staining.
  • In the two-step cold method, the specimen is first treated with the primary acid-fast stain, allowing the dye to penetrate the bacterial cell wall without the use of heat. The specimen is then subjected to decolorization and counterstaining in single Step according to the kit procedure. Acid-fast organisms retain the primary stain, whereas non-acid-fast material is decolorized and simultaneously takes up the counterstain.
  • Under light microscopy, acid-fast bacilli appear as distinct red to pink-red slender rods against a contrasting background.
