- Gill II Hematoxylin is the most widely used H&E Hematoxylin in modern histology labs worldwide. It is double the strength of Gill I and formulated as per Dr. G. W. Gill’s formula.
- It is a progressive, aluminum-based, mercuric oxide-free nuclear stain. It is the perfect balance – Strong enough to stain all tissues (Liver, Colon, Breast, Prostate) in just 2 minutes, but light enough to still be progressive (No differentiation needed).
- If Gill I is for Cytology, Gill II is for Histology.
- 99% of NABL accredited labs and large corporate pathology chains in India have now shifted from Harris to Gill II because Harris causes over-staining, metallic scum, and batch-to-batch variation.
- Perfect Strength for H&E: Contains 4.0g/L Hematoxylin – the optimal concentration. Gill I is too light for histology (needs 4 min), Gill III is too dark (risk of opaque nuclei). Gill II is the sweet spot.
- True Progressive – No Acid Alcohol Needed: Eliminates differentiation step completely. Saves cost of Acid-Alcohol, saves technician time, and eliminates 90% of H&E errors (faded nuclei due to over-differentiation).
- Zero Metallic Scum & Precipitate: Formulated with high Ethylene Glycol (250ml/L) and stable oxidizer. Unlike Harris which forms black metallic film on stain jar and black dots on slides, Gill II is clean till last drop. No daily filtering.
- Crystal Clear Nuclear Chromatin: Gives transparent staining – pathologists can see chromatin pattern, nucleoli, mitosis, nuclear grooves very clearly. Critical for cancer grading (Breast, Prostate). Harris gives opaque black nuclei.
- Mercury-Free & Eco-Friendly: Contains Sodium Iodate as oxidizer, not Mercuric Oxide. Safe for technicians and for disposal. Complies with NABL & Pollution Control norms.
- 2-Year Shelf Life: Chemically ripened formula is ultra-stable. Harris expires in 6-8 months. Gill II is stable for 24 months at Room Temperature. No loss of staining intensity.
Double-Strength Progressive Lake
Same mechanism as Gill I, but with double dye concentration for faster histology workflow.

- Step 1 – Lake Formation: 4g/L Hematein (oxidized Hematoxylin) + Aluminum Sulfate forms intense Aluminum-Hematein Lake.
- Step 2 – Progressive Saturation:This cationic lake binds to anionic DNA phosphate groups in nucleus. Because concentration is double of Gill I, saturation is achieved in 90 seconds to 2 minutes for 4-micron sections.
- Step 3 – Self-Limiting: Once all DNA binding sites are occupied, no further dye uptake occurs. So even if technician leaves slide for 4 minutes, it will NOT over-stain. This is the biggest advantage over Harris.
- Step 4 – Blueing: Simple tap water wash at pH >7 converts red-purple lake to deep blue-purple in nucleus.
