- Leishman staining is based on the Romanowsky staining principle, using acidic and basic dye components to produce differential staining of cellular structures.
- The basic dye component interacts predominantly with acidic cellular structures such as nuclear chromatin, while the acidic dye component stains basic cytoplasmic components. The interaction of these dye components produces characteristic blue-purple nuclear staining and varying shades of pink, lilac and purple in cytoplasmic and other cellular structures.
- This differential staining provides contrast that allows individual blood-cell types and their morphological features to be examined microscopically.
