Acetone
- IHC Compatible Acetone is a premium, molecular-biology grade, anhydrous dehydrant and precipitating fixative, specially purified for Immunohistochemistry (IHC), Immunofluorescence (IF), and Frozen Sections.
- Unlike regular lab-grade acetone that contains water, aldehydes, and acidic residues, IHC grade is double-distilled, water-free (<0.1% moisture), pH neutral, and DNase/RNase free. It fixes tissue by rapid protein coagulation WITHOUT cross-linking, thus it preserves the most sensitive epitopes and antigens that formalin destroys.
- It is the first-choice fixative for Lymphoma panels, Intra-operative frozen sections, and research antibodies.

1. Acetone rapidly removes lipids and dehydrates tissue in 5-10 minutes at -20°C to 4°C. It coagulates and precipitates proteins in their native state. Cell morphology is locked without chemical alteration. No cross-linking = 100% antigenicity preserved.
2. Protocol (Cryostat): Fresh / Unfixed Tissue → Cut at 4-6µm → Immerse in chilled Acetone (-20°C) for 5-10 min → Air Dry 2 min → Proceed to IHC/IF
3. Protocol (Cell Lines / Cytology): Cytospin / Smear → Air dry → Cold Acetone fix 5 min.

In some fatty biopsies (Breast, Brain, Lipoma) and rapid processing protocols, Acetone acts as an intermediate dehydrant. It is fully miscible with ethanol, xylene, and paraffin. It removes fat faster than ethanol and clears better than IPA.
