Congo Red Stain Kit is a histopathology staining system designed for the demonstration and identification of amyloid deposits in tissue sections. Congo Red binds selectively to amyloid and produces a characteristic red to deep pink staining under bright-field microscopy. When examined using polarized light, Congo Red–stained amyloid demonstrates characteristic birefringent/anomalous optical colours, classically described as apple-green birefringence.

Congo Red dye binds to the organized fibrillar structure of amyloid deposits. The bound dye molecules become aligned along the amyloid fibrils, producing characteristic optical properties when viewed between crossed polarizers. Amyloid deposits can therefore be assessed by combining their Congo Red staining characteristics with polarized-light examination.

Microscopy Under ordinary light microscopy, amyloid typically appears red to deep pink following Congo Red staining. Under appropriately configured polarized-light microscopy, Congo Red–stained amyloid may show characteristic green and other anomalous birefringent colours.
