Standard Bone Marrow Fixative
It is the Gold Standard and NABL/CAP/WHO recommended fixative for Bone Marrow Trephine Biopsy. It provides optimal preservation of marrow architecture, cellular morphology, and trabecular bone matrix while allowing perfect decalcification and immunohistochemistry (IHC).
- Standard Bone Marrow Fixative – IHC Compatible Prevents formation of Formalin pigment (acid hematin) that mimics parasites in marrow.
- Isotonic Formulation: Phosphate buffer maintains osmolarity ∼300 mOsm – No shrinkage or swelling of marrow cells.

- Optimal Penetration: Penetrates dense cortical bone at 1mm/hour – Ideal for 1.5-2cm trephine core.

1. Fixation :
- Penetration Phase (0-2 hrs): It rapidly diffuses into marrow core and fixes outer hematopoietic cells.
- Cross-Linking Phase (2-12 hrs): Forms covalent methylene bridges between lysine residues of proteins – locks cellular morphology and prevents autolysis.
- Stabilization Phase (12-24 hrs): Complete fixation of central marrow fat and bony trabeculae – prepares tissue for Standard & Rapid – IHC Compatible decalcification without DNA damage.
