PAS-D Stain Kit
PAS-D Stain Kit is a specialized histopathology staining system combining Periodic Acid–Schiff (PAS) staining with diastase digestion. It is used to demonstrate PAS-positive, diastase-resistant substances in tissue sections and is particularly useful for differentiating glycogen from other PAS-positive materials.

The PAS-D technique consists of two main stages:
- Diastase digestion: Tissue sections are treated with diastase to enzymatically remove glycogen.
- PAS staining: Periodic acid oxidizes suitable tissue carbohydrates, producing aldehyde groups that react with Schiff reagent to generate a characteristic magenta coloration.
- Comparison of the PAS and PAS-D reactions: helps determine whether a PAS-positive substance is diastase-sensitive (such as glycogen) or diastase-resistant.
- Helps differentiate glycogen from diastase-resistant PAS-positive substances.
- Enhances the diagnostic utility of conventional PAS staining.
- Provides clear visualization of carbohydrate-rich tissue components
