  • Modified Mayer’s Hematoxylin is an alcohol-free, mercury-free, progressive nuclear counterstain specially modified for Immunohistochemistry (IHC), Special Stains, and Routine H&E.
  • Original Mayer’s formula (1903) was too weak and unstable. We have modified it by increasing Hematoxylin concentration and adding stabilizers to give a stronger, stable stain that gives light blue transparent nuclei – without masking the brown DAB signal of IHC.
  • This is the #1 Counterstain used worldwide for IHC – Recommended in all DAKO, Ventana, Leica, Biocare IHC kits.
  • Why Every IHC Lab Needs Modified Mayer’s ?

Gentle Progressive Counterstaining

  • Step 1 – Pure Aqueous Lake: Hematein + Aluminum Ammonium Sulfate forms a light, aqueous Aluminum-Hematein lake. No alcohol or ethylene glycol, so it never interferes with IHC antigen-antibody-DAB complex.
  • Step 2 – Light Progressive Binding: Low concentration (1g/L Hematoxylin) binds slowly and lightly to nuclear DNA. It stains only chromatin, not over-stains.
  • Step 3 – Perfect Contrast with DAB: In IHC, DAB gives brown cytoplasmic / nuclear signal. If counterstain is dark blue, brown + dark blue = black, difficult to read. Modified Mayer’s light blue + brown = excellent contrast. Pathologist can easily score ER/PR intensity and %.
  • Step 4 – No Differentiation Needed: Progressive stain – washes out cleanly in tap water, no acid-alcohol needed, so delicate IHC tissue never detaches.
