Toluidine Blue Stain Kit is a special histological stain based on Toluidine Blue O for the demonstration of mast cells, acidic mucopolysaccharides, mucins, cartilage and other basophilic tissue components. Its characteristic metachromatic reaction allows mast-cell granules and other highly acidic tissue substances to develop a colour different from the original blue dye.

Toluidine Blue is a basic metachromatic dye that binds to acidic tissue components. When the dye interacts with highly concentrated anionic groups, such as those present in mast-cell granules containing heparin, dye molecules aggregate and undergo a characteristic metachromatic colour shift. Consequently, the mast-cell granules appear violet, purple or red-purple, while many other tissue components remain blue.
