New Methylene Blue (NMB) Stain Kit is a supravital staining system designed for the microscopic demonstration and enumeration of reticulocytes and for the detection of Heinz bodies and selected erythrocyte inclusions. It highlights residual ribosomal RNA in immature red blood cells as blue reticulofilamentous material, allowing reticulocytes to be differentiated from mature erythrocytes.

New Methylene Blue is a supravital dye that is applied to unfixed blood cells. It interacts with residual ribosomal RNA and other reticulofilamentous material present in immature red blood cells. Following incubation, the residual RNA becomes visible as blue granular or filamentous material, while mature erythrocytes, which have lost their residual RNA, show little or no reticulofilamentous staining.
The stained preparation can be examined microscopically and reticulocytes can be expressed as a percentage of the red-cell population. NMB may also demonstrate Heinz bodies, which appear as pale blue intracellular inclusions, although dedicated Heinz-body stains such as methyl violet may provide stronger visualization when specifically required.

Supports microscopic examination of selected red-cell inclusions, including Heinz bodies and HbH-related inclusions under appropriate staining conditions.
