  • Modified Gram Stain Kit is a differential staining system designed for the demonstration and differentiation of Gram-positive and Gram-negative bacteria in direct smears and body-fluid specimens. The modified staining procedure provides good bacterial contrast against the cellular and specimen background, supporting microscopic evaluation of bacterial morphology, distribution and Gram reaction.

  • Gram staining is based on the differential retention of a crystal violet–iodine complex by bacterial cell walls. Following primary staining and iodine treatment, the decolorization step differentiates bacteria according to their cell-wall characteristics.
  • Gram-positive bacteria generally retain the primary dye complex and appear blue to violet/purple, while Gram-negative bacteria are decolorized and subsequently take up the counterstain, appearing pink to red.
  • The modified staining sequence is optimized for smear and body-fluid preparations to provide clear bacterial visualization while maintaining sufficient contrast with the surrounding cellular material.
