1. Fixation & Hematoxylin: Nuclei become Blue.
2. Differentiation: Excess blue removed.
3. Our Eosin Y 1% Alcoholic: Enters cytoplasm. As an anionic dye, it binds to cationic proteins in cytoplasm, collagen, muscle, RBCs.
4. Dehydration & Mounting: Pink becomes permanent and transparent.
