IHC Compatible Sulfur-Free Xylene is the gold standard clearing agent, specially refined for modern biopsy tissue processing where IHC, IF, and Molecular preservation is critical.
Standard industrial xylene contains trace sulfur compounds (thiophene, mercaptans) from petroleum refining. These residues cause non-specific background staining, mask epitopes, reduce signal-to-noise ratio in IHC, and cause tissue hardening.
Sulfur-Free grade is triple-distilled, thiophene-free isomeric mixture (ortho-, meta-, para-) with purity >99.5%. It is the most efficient clearing agent in a histology lab.

Clearing in Tissue Processing :

After graded Alcohol dehydration, tissue still contains alcohol. Xylene is miscible with BOTH Alcohol and paraffin. It displaces Alcohol completely and renders the tissue transparent (hence the name “clearing”). This transparency confirms complete dehydration. It then allows molten paraffin to infiltrate fully for perfect embedding.

Deparaffinization in Staining :

Before H&E or IHC staining, paraffin must be removed. Sulfur free xylene rapidly dissolves wax from 4-micron sections in 2 x 5 min, without leaving residue. This allows rehydration and uniform stain uptake.

Coverslipping :

Final step after dehydration (95% → 100% Alcohol→ Xylene) gives high refractive index for DPX mounting. Gives brilliant nuclear/cytoplasmic clarity.

For Pathologists :

Proper hardness of blocks, easy cutting of 2-3 µm consecutive ribbons, no cell shrinkage, clear definition of nucleus & cytoplasm, intact N:C ratio, crisp H&E.

For IHC Lab :

Validated with 50+ antibodies – Vimentin, p63, PSA, Pan-CK, CD3, CD20, ER, PR, SMA, Thyroglobulin. Identical SNR vs reference, very low background. Compatible with IHC, ISH, and Multiplex IF.
