Scott’s Tap Water Substitute is a controlled alkaline bluing reagent designed for routine Hematoxylin & Eosin (H&E) staining in histopathology and cytology. It converts the hematoxylin-stained nuclei from a reddish-purple tone to a crisp blue to blue-black color, enhancing nuclear contrast and definition.

Promotes conversion of hematoxylin to its characteristic blue nuclear color.
Provides clear nuclear chromatin and membrane definition.

After hematoxylin staining and differentiation, nuclear staining may appear red-purple or reddish-brown. Scott’s Tap Water Substitute provides an alkaline environment that promotes the conversion of the hematoxylin staining complex to the desired blue/blue-black form. The section is subsequently rinsed and proceeds to the eosin counterstaining step.
