Masson’s Trichrome Stain Kit is a special histological staining system designed for the differential demonstration of collagen fibres, muscle fibres, connective tissue and nuclei in tissue sections. It provides strong colour contrast between collagen and muscle, making it particularly useful for evaluating connective-tissue architecture, fibrosis and tissue remodelling.
The stain is widely used in histopathology for liver, kidney, heart, lung, skin, gastrointestinal and other tissue specimens where differentiation of collagen from muscle and cellular components is required.

Masson’s Trichrome staining is based on the differential binding and penetration of acidic dyes within tissue components. The section is first stained with Weigert’s iron hematoxylin to demonstrate nuclei. An acidic red dye system then stains muscle, cytoplasm and other acidophilic tissue components.
Treatment with phosphomolybdic/phosphotungstic acid differentiates the red dye from collagen and prepares collagen fibres for the subsequent fibre stain. Aniline blue then selectively demonstrates collagen, producing a contrasting blue colour. A final acetic-acid step helps improve colour differentiation and clarity.
