  • Ziehl–Neelsen (ZN) Stain Kit is a conventional hot staining system designed for the microscopic demonstration of acid-fast bacilli (AFB) in clinical specimens. It is widely used for the detection of mycobacteria in appropriately prepared smears, particularly in specimens where mycobacterial infection is suspected.
  • The method uses heated carbol fuchsin as the primary stain, followed by acid-alcohol decolorization and a contrasting counterstain. Acid-fast organisms retain the primary stain and appear distinctly red against a contrasting background.

  • Acid-fast bacilli possess a cell wall containing a high concentration of mycolic acids and other lipid-rich components, which makes them resistant to decolorization by acid-alcohol after penetration of the primary stain.
  • In the hot Ziehl–Neelsen method, strong carbol fuchsin is applied to the smear and gently heated to facilitate penetration of the stain into the waxy bacterial cell wall. After cooling, the smear is treated with an acid-alcohol decolorizer. Acid-fast organisms retain carbol fuchsin, whereas non-acid-fast material is decolorized.
  • A counterstain is then applied to provide background contrast. Under light microscopy, acid-fast bacilli appear bright red to pink/red rods against a contrasting blue or otherwise counterstained background.
