Myeloperoxidase (MPO) Stain Kit is a cytochemical staining system designed for the demonstration of myeloperoxidase activity in blood and bone marrow smears. MPO is a characteristic enzyme of cells of the myeloid lineage and is particularly useful in the cytochemical evaluation and classification of leukemic and other hematological disorders.

The MPO reaction is based on the enzymatic activity of myeloperoxidase present in the cytoplasmic granules of myeloid cells. In the presence of an appropriate peroxide substrate and chromogenic reagent, MPO catalyzes an oxidation reaction that produces an insoluble coloured reaction product at sites of enzyme activity. The resulting cytoplasmic staining allows MPO-positive cells to be identified microscopically.

Interpretation

MPO-positive cells demonstrate a characteristic coloured cytoplasmic reaction, particularly in cells containing myeloid granules. The staining result should be interpreted together with peripheral blood/bone marrow morphology and other laboratory findings rather than used as a standalone diagnostic test.
